Introductory evaluation of somatic hybrids Solanum pinnatisectum + Solanum tuberosum cv. Keřkovské rohlíčky in terms of morphology and resistance to Phytophthora infestans (A)

Interspecific somatic hybrids of wild species S. pinnatisectum PI320342 (2n = 2x = 24, 1EBN) and regional cultivar S. tuberosum Keřkovské rohlíčky (2n = 4s = 48, 4EBN) were evaluated in terms of ability for acclimatization, plant morphology, tuberization, and resistance to Phytophthora infestans in an in vitro test. The percentage of affected leaf area was used for evaluation (AUDPC). All somatic hybrids employed in this study were hexaploids (2n = 6x = 72), all were acclimatized and tuber bearing. In vitro test of resistance to PI showed strong resistance of wild parent and susceptibility of cultivated parent. Somatic hybrids varied between parental genotypes: one of them showed high level of resistance while two displayed susceptibility, the remaining had an intermediate character. Somatic hybrids seem to be promising breeding material.

Molecular probing of Potato Virus S genome (A)

Complete genomes of three isolates of Potato virus S (PVS) were cloned and sequenced. The complete sequence of PVS genome enabled to develop an immunocapture RT-PCR probing of the PVS genome. Using this system, the sequence variability of 11 genome zones was examined for 34 PVS isolates including 15 PVS-CS variants that caused a systemic infection in Chenopodium quinoa. A broad variability between PVS isolates and diverse sequence variants was found. cDNA fragments covering the coat protein (CP) leader and CP-coding region (approx. 420 bp) were pooled for PVS-O and Chenopodium-systemic PVS isolates (PVS-CS) and corresponding cDNA libraries were screened for sequence variants. Genome probing suggested that the PVS-CS isolates analyzed were close to the ordinary European isolates of ordinary strain of PVS (PVS-O) but distant to the original Andean strain of PVS (PVS-A).

Origin and development of new genotypes using MAS (marker assisted selection)

We managed to obtain characterized hybrid materials using wild species of the Solanum genus, which have a large amount of genetic diversity for economically important traits. These materials will be used next year for testing for selected properties, for morphological evaluation and for further backcrossing. Prospective materials will be transferred to the gene bank of the Potato Research Institute in Havlíčkův Brod.

Occurrence of silver scurf (Helminthosporium solani) on potato tubers, in soil and during storage in the years 2017 and 2018

Silver scurf infection of potato tubers was studied for several varieties in exact field trials in connection with harvest dates between the years 2016 and 2018 and further in two potato stores during storage season 2018/2019. The number of infected tubers and silver scurf severity significantly increased with later harvest date. An increase in silver scurf occurrence was also determined during storage period. This trend was clear in all studied years and all varieties. Considering the fact that silvers scurf is an important disease negatively impacting internal quality of ware potatoes and that direct fungicide control is currently not real, the selected harvest date could be an important preventive measure against this pathogen.
